treatments to noise-induced hearing loss

Study proves possible drug treatment for noise-induced
Hearing loss

Heidelberg/New York, 30. June 2010

Scientists from the University of Auckland in New Zealand have found, that the hearing in rats after noise-induced hearing loss with an effective, new drug can be restored. was found at the groundbreaking discovery, which are induced by the injection of so-called drug "ADAC" adenosine receptors activated in cochlear tissue, leading to a recovery of hearing. The findings pave the way for effective, non-surgical treatment methods for recovery of hearing after noise pollution-induced hearing loss. The work of Dr. Srdjan Vlajkovic and his Team1 appears in the special-task on the inner ear of the journal Purinergic Signalling by Springer.

Noise-induced hearing loss is one of the most common occupational diseases, threatened with up to five percent of the world's population are. The disease is particularly widespread in the military and in industry (Construc-sen, Mining, Forestry and aviation industry). Currently, hearing aids and cochlear implants are the only treatments for hearing loss. Only recently have drug Therapiekon concepts are proposed for noise-induced hearing loss, because until now there are practically no methods of treatment, with damage to the inner ear and the fixed effects of hearing loss can be reduced.

In the study by Dr. Vlajkovic and his team is the treatment potential of adenosine amine congeneric (ADAC) – a selective A1 adenosine receptor agonists – investigated in noise-induced hearing loss. Here were Wistar rats for in an acoustic chamber 2 – 24 Hours exposed to noises Schmalbandge-, bring to cochlear damage and permanent hearing loss. Via an injection into the abdomen, the animals were ADAC or a placebo formulation administered, either as a single injection after six hours or more injections a day. Using the so-called specific reaction associated with the brainstem (ABR) scientists study the hearing of the rats before and after treatments. In addition, they used a histological techniques, to determine the number of missing hair cells after noise exposure and noise-induced production of free radicals.

Their study results show, that cochlear damage and hearing loss in rats, the narrow-band noises were exposed, by the administration of ADAC can be significantly reduced. The best results were obtained with early administration of ADAC made six hours after noise exposure, compared to a later start treatment after 24 Hours. The most sustainable treatment strategy was the repeated injection of ADAC for the duration of five days after noise exposure. By this method of treatment of hearing loss was reduced significantly and improved the preservation of hair cells significantly.

The authors come to the conclusion: "This scientific study of the important role of adenosine signaling underscores the relief of cochlear damage, the oxidative stress caused. In particular, the ADAC has emerged as an attractive pharmacological agent for the treatment of noise-induced cochlear damage in both acute and prolonged noise exposure. "
